I don't really have any reason to suspect something went wrong in the update but a loss of 161MB seems excessive. What package got shrunk so dramatically?

My guess would be it was something mesa-related.

If this is new, then it is most likely the reason.
edit: To show the difference:
Some time ago I built my lib32-mesa with "--build=x86_64-pc-linux-gnu --host=i686-pc-linux-gnu" because there was some bug with crosscompiling. Unfortunately the configure script doesn't support --with-llvm-shared-libs with this sort of crosscompiling but I saw in arch's lib32-mesa PKGBUILD that it apparently builds again with the old style cross compile, so I did it:
-250,21 MiB on my own lib32 mesa package.

I don't thing so. I update arch every ~ 2 days and about 2 weeks ago most of updated packages are smaller then before. Not only mesa (but here the difference is very noticeable), but most of packages eg. libreoffice too. I haven't made any statistics, but it is how I can remember.

No, the correct reason has been highlighted above. llvm shared libs has dramatically reduced the number of copies in the mesa page which was previously at 7.
