When do I set the root password

I've been using linux for a couple years now, so I feel kind of stupid  asking, but when are you supposed to set the root password? After the install, I booted into single user mode, but when I tried to change it then, it said it couldn't lock the passwd file. Well, thanks for any help.

May the Schwartz be with you.
