After several days of trying different things, I got a Zune to sync via VirtualBox without having to create any new NTFS partitions or anything crazy like that. Other than the VM, my environment was kept 100% native (as in, I didn't have to change anything aside from creating a few files here and there). Basically I found an ext2 windows driver that worked and was good to go.
So, what I'm wondering is.... would this be something fitting of throwing on the wiki? I'll write it up if people are interested.

I'd be interested in something, as I'm currently trying the same thing. So you're not using the built-in file sharing in VB (i.e., mount as network drive)? How do you get XP in the VM to recognize your ext2/ext3 partition? I installed and ext3 driver and generated a vmdk but it didn't work. Is this what you did?
My Zune software sees the files, and I can play them fine. My only problem is with syncing. So far, I have only been able to sync one picture and one song - on separate occasions. The rest of the songs fail with "Can't access the file". Did you do anything specific to get syncing working?
By the way, I do think that such a topic would be appropriate for the wiki.

Are you getting that error while using the mounted network share? This is why I had to create rawdisk/writethrough .vmdk files, because I got that same error when I used the shared folders.
Here's what I did, in a nutshell:
1. Created a writethrough image (described in section 9.9 of the VirtualBox manual) of the partition I keep my music on.
2. Added that image to the VM.
3. Booted up the VM.
4. Installed an ext2 driver in the XP VM.
5. Added the folder to the Zune software and synced.
The secret was finding an ext2 driver that worked properly. I've used a program called Ext2IFS in the past, and had mixed luck with it, but thanks to the links on this page (scroll down to "Some Other Drivers"), I found Ext2fsd, which, so far, has worked with every partition I've tried it with.

Hi!
I'm having the same problems trying syncing my Zune via VirtualBox, I can add and, even, play my music from the network drive but when I try to sync the Zune software shows me an error like "Can't access the server".
I tried to follow the "tonyisnt" instructions but in the first step, when I try to create a Writeghrough image and add it to my current virtual Windows XP, VirtualBox shows me an error related with access permissions.
My music is allocated in a FAT32 partition, what can I do?
