After upgrading to 334.21, there is no cuda option in the user preference of blender and blender prints
CUDA cuInit: Unknown error
to the terminal on startup.
I've looked at this bug report: https://bugs.archlinux.org/task/39141, but the issue persists even after upgrading to nvidia 334.21-2.

According to some reports, running something (anything) that uses CUDA as root first, then running the CUDA-using app as a regular user works. Have you tried that?

I have the same problem.... after running:
primusrun blender i get CUDA cuInit: Unknown error
By the way, with optirun just doesn't works, simply doesn't show me any other card than CPU
any ideas?

Did you try first with sudo?
sudo primusrun blender
Then try with your regular user.

Just FYI CUDA appeared as compute option after I ran blender as root. I was getting the same error as OP before.

Same for me. Running as root first then as user solved the problem for the time being.
